Huperzine-A (from Huperzia Serrata)
Updated October 11, 2026 · By Sophie Callahan
The Short Answer
Huperzine-A is a sesquiterpene alkaloid extracted from Chinese club moss (Huperzia serrata) that inhibits acetylcholinesterase, the enzyme that breaks down acetylcholine in the brain. This increases available acetylcholine, supporting memory and learning. Unlike most nootropic botanicals that require hundreds of milligrams, Huperzine-A is effective at microgram-level doses (200–400 mcg/day), making it one of the most potent natural cognitive support compounds by weight. Published RCTs date back to 1995.
What is Huperzine-A (from Huperzia Serrata)?
Huperzine-A is a naturally occurring sesquiterpene alkaloid isolated from the firmoss Huperzia serrata (also known as Chinese club moss or Qian Ceng Ta). It has been used in traditional Chinese medicine for centuries for memory complaints and inflammation. Modern supplements extract and purify Huperzine-A to a standardized concentration, typically 1% of the raw plant material. What distinguishes Huperzine-A from most botanical nootropics is its potency: effective doses are measured in micrograms (200–400 mcg), not milligrams.
How it works
Huperzine-A's primary mechanism is reversible inhibition of acetylcholinesterase (AChE), the enzyme that degrades the neurotransmitter acetylcholine in the synaptic cleft. By slowing acetylcholine breakdown, Huperzine-A increases the concentration and duration of acetylcholine signaling at cholinergic synapses — the neural pathways most directly involved in memory formation, attention, and learning. This mechanism is pharmacologically similar to prescription acetylcholinesterase inhibitors like donepezil, though Huperzine-A has a shorter duration of action and different binding characteristics. Secondary mechanisms include protection against glutamate-induced neurotoxicity and potential antioxidant activity in neural tissue.
What the evidence says
Huperzine-A has been studied primarily in populations with cognitive decline, including Alzheimer's disease and vascular dementia. The evidence base includes multicenter RCTs and a Phase II trial for traumatic brain injury recovery.
- Huperzine-A at 200 mcg twice daily improved memory and cognitive function in Alzheimer's patients. — A multicenter, prospective, double-blind, parallel, placebo-controlled, randomized trial evaluating efficacy and safety of Huperzine-A tablets (0.2 mg) in patients with Alzheimer's disease. (Xu et al., 1995 (Multicenter RCT))
- Huperzine-A was explored as a treatment for cognitive deficits following moderate-to-severe traumatic brain injury. — A Phase II randomized, double-blind, placebo-controlled pilot study investigating Huperzine-A for cognitive, mood, and functional recovery after moderate-to-severe TBI. (Zafonte et al., 2020 (Phase II RCT))
Effective dosage
Clinical trials have used 200–400 mcg/day of purified Huperzine-A. A 1995 multicenter RCT by Xu et al. used 200 mcg twice daily (400 mcg total) in Alzheimer's patients. Because Huperzine-A is effective at microgram levels, even small allocations within proprietary blends can potentially deliver a clinically relevant dose — a key distinction from most botanical nootropics. It can be taken with or without food. Cycling (e.g., 2–4 weeks on, 1 week off) is sometimes recommended by practitioners to prevent receptor downregulation, though this is based on clinical judgment rather than published trial data.
Safety and interactions
Huperzine-A is generally well tolerated at doses up to 400 mcg/day in clinical trials. Common side effects include nausea, diarrhea, and dizziness, typically at the higher end of the dose range. Because it inhibits acetylcholinesterase, it should not be combined with prescription acetylcholinesterase inhibitors (donepezil, rivastigmine, galantamine) without medical supervision — the combined cholinergic effect can cause excessive acetylcholine accumulation. It may also interact with anticholinergic medications and beta-blockers. The vendor warning on Marobrain about blood thinners and alpha-blockers is appropriate for this ingredient.
